Common Approaches of Cytochrome P450 (CYP) Induction Assays

چکیده

The induction of enzymes is a defensive mechanism for some xenobiotics, but it may alter the drug's safety and efficacy by altering activity metabolic enzymes. One major families involved in phase I metabolism Cytochrome P450 (CYP) which get induced certain drugs. Concomitant administration drugs due to chronic disease or polypharmacy, inducers among them cause toxicity reduce plasma concentration at sub-therapeutic level. This one dangerous types drug-drug interactions, predictable & preventable. CYPs three nuclear receptors, including aryl hydrocarbon receptor (AhR); constitutive androstane (CAR); pregnane X (PXR). Without identification during drug development, enzyme phenomenon new molecule noticed only pharmacovigilance. Though, this CYP not be barrier possible DDI treatment failure. According FDA guidelines, pharmaceutical industries adopted In-vitro, Ex-vivo In-vivo techniques based on different developmental stages. results are also interpreted regulatory bodies guidelines. For In-vitro assay best accepted method using primary hepatocytes either fresh cryopreserved, liver slices species in-vivo, clinical investigations extreme option. paper reviews current industry approaches assays evaluate potentiality as an inducer.

Modeling of interactions between xenobiotics and cytochrome P450 (CYP) enzymes

The adverse effects to humans and environment of only few chemicals are well known. Absorption, distribution, metabolism, and excretion (ADME) are the steps of pharmaco/toxicokinetics that determine the internal dose of chemicals to which the organism is exposed. Of all the xenobiotic-metabolizing enzymes, the cytochrome P450 (CYP) enzymes are the most important due to their abundance and versa...

Effective cytochrome P450 (CYP) inhibitors isolated from tarragon (Artemisia dracunculus).

Two effective cytochrome P450 (CYP) inhibitors were isolated from tarragon, Artemisia dracunculus. Their structures were spectroscopically identified as 2E,4E-undeca-2,4-diene-8,10-diynoic acid isobutylamide (1) and 2E,4E-undeca-2,4-diene-8,10-diynoic acid piperidide (2). Both compounds had dose-dependent inhibitory effects on CYP3A4 activity with IC50 values of 10.0 ± 1.3 µM for compound 1 and...

Cytochrome P450 expression (CYP) in non-small cell lung cancer.

The cytochrome P450 (CYP) is associated with tumor development and progression as well as activation of anti-cancer prodrugs and their metabolic clearance. In this study, we investigated the expression of aryl-hydrocarbon receptor (AH-R) and four CYPs (CYP1A1, CYP2A6, CYP2E1 and CYP3A) as putative diagnostic markers in 78 non-small cell lung cancers (NSCLC) along with clinical features of the p...

Validated assays for human cytochrome P450 activities.

The measurement of the effect of new chemical entities on human cytochrome P450 marker activities using in vitro experimentation represents an important experimental approach in drug development. In vitro drug interaction data can be used in guiding the design of clinical drug interaction studies, or, when no effect is observed in vitro, the data can be used in place of an in vivo study to clai...
